Microbiology

Fluxion's BioFlux System supports a wide variety of applications in microbiology, from high content analysis to antimicrobial screening. Each microfluidic channel serves as an adaptable experimental chamber ready to be used for physiologically-relevant shear flow assays. The BioFlux system is especially suited for biofilm analysis under shear flow as well as targeted antimicrobial development against biofilms. Other key applications include bacterial adhesion and host-pathogen interactions using a variety of substrates and mammalian cell monolayers. BioFlux enables in vivo relevance for your microbiology experiments in a convenient, high throughput and low volume well-plate format.

Representitive Examples of Microbiology Assays Using BioFlux
 Viability staining

Pseudomonas fluorescens biofilm grown under 2.0 dyn/cm2 controlled shear flow for 24 hours. Stained with BacLight viability stain (Invitrogen).

Imaged with QICAM, Nikon TS100 and Mercury Lamp.
 Biofilm morphology

Pseudomonas aeruginosa PAK biofilms grown for 24 hours in the BioFlux microfluidic channels. Side by side biofilms (top) before disruption and side by side biofilms (bottom) one disrupted and one intact.

Phase contrast imaging with Nikon Elcipse TS100 and QICAM.
 Host-pathogen interactions under flow conditions

Pseudomonas aeruginosa PAK - GFP attach to Calu-3 cells. Image was capured under shear flow. Fluorescent bacteria were imaged and then overlayed on the phase contrast micrograph. Arrows indicate bacteria firmly attached to host cells.
 High-throughput screening

Pseudomonas aeruginosa biofilms were grown in the serpentine regions of BioFlux plates, exposed to biocides and then stained with Live/Dead reagents (Invitrogen). Plates were then scanned using a microplate fluorimeter. Shown is an early image of bacteria stained with Syto9, the live component of the BacLight Viability staining kit.
