Automated Electrophysiology

Fluxion's IonFlux system provides automated patch clamping for recording from ligand-gated and voltage-gated ion channels. A wide variety of electrophysiology applications are supported, including ion channel drug screening and pharmacology.

Representative Examples of Automated Electrophysiology Experiments Using IonFlux

GABA-A channel recording

Continuous recording from an ensemble of CHO cells expressing GABA-A receptors during successive application of increasing GABA concentrations is shown. The reported EC50 is 3µM.
 Kv2.1 Currents During Perfusion

Responses to two TEA (tetraethylammonium) concentrations are shown in two different cells. Voltage step protocol eliciting a full I-V curve was performed during steady state TEA perfusion. 15-30% inhibition was observed at 10mM TEA, while almost complete inhibition was observed at 100mM TEA. Post-perfusion measurements reflect slow inactivation of the Kv2.1 channel over time.
 P2X3

Agonist evoked inward currents from two different RLE cells during exposure to αβ methylene(Me) ATP in RLE cells stably transfected with hP2X3 receptor/channels.
 TRPV1

Membrane current recorded in response to 50µM capsaicin applied for 5s to a single CHO – hTRPV1 cell at one trapping site in an IonFlux 100 device. Normalized dose-response curve based on recordings similar to the left panel.
